 TRIZIVIR

Approved for use in HIV infection in combination with other antiviral drugs.

DESCRIPTION: Trizivir® is a combination of three NRTIs: abacavir, 3TC (lamivudine) and AZT (zidovudine).

DOSE: One tablet twice a day. Each tab. contains 300 mg ABC, 150 mg 3TC and 300 mg AZT. Because of the fixed dosage , people weighing > 40 kg or with renal impairment should not take Trizivir.

SIDE EFFECTS: The most common side effects of Trizivir are the same as with Retrovir (AZT), Epivir (3TC), and abacavir (Ziagen). They include headache, upset stomach, nausea, and fatigue. See ABC, 3TC and AZT for listings. Trizivir is associated with fatal hypersensitivity reactions. Enlarged fatty liver and lactic acidosis, including fatal cases have been reported with the NARTIs.

RESISTANCE: See ABC, 3TC and AZT

DRUG INTERACTIONS: Trizivir should not be taken with ddC (zalcitabine, Hivid®) or with d4T (stavudine, Zerit®). Blood levels of 3TC may be increased by septrin.

See ABC, 3TC and AZT

ADVANTAGES

DISADVANTAGES

Simple regimen – low tablet numbers

Maybe less potent than PI or NNRTI containing combination

Appear to be associated with decreases in triglycerides and cholesterol

Hypersensitivity problems with ABC

Saves PI and NNRTI options

Possibility of increased NARTI toxicity with 3 NARTIs – mitochondrial toxicity
